Studies on the Activity of Microsomal Alkaline Phosphatase of the Rat Kidney

Abstract
|
|
|
The effect of steroid hormone and various agents on the alkaline phosphatase activity of the microsomal fraction of rat kidney was studied in vitro.
The alkaline phosphatase activity was measured by method described the Bessey et al. using p-nitrophe¡©nylphosphate disodium as substrate. The results were summarized as follows.
1. Renal alkaline phosphatase activity was augmented markedly by prednisolone, but slightly augmented by cortisone.
2. Cysteine and phenylalanine repressed renal alkaline phosphatase activity while cystine, glutathione, histidine or tryptophan had little influence on the alkaline phosphatase activity.
3. Increasing the osmolality by the addition of substance such as sucrose or NaCl in the medium resu¡© Red in repression of activity, in the following order; LiCl>NaCI>KCI, Sucrose>Dextrose>Urea.
4. Diamox repressed markedly renal alkaline phosphatase.
5. Zinc chloride augmented markedly on renal alkaline phosphatase activity but sodium sulfate had little effect.
